An Oroboros O2k high-resolution respirometry unit from Oroboros Instruments GmbH. This unit is used for studying mitochondrial physiology, including its application with isolated mitochondria, cells (intact or permeabilized), and permeabilized tissues. The instrument measures both oxygen concentration and flow with very high precision in real time, allowing the injection of various compounds (such as substrates and/or mitochondrial inhibitors) using Hamilton microsyringes. The instrument features two respiration chambers with minimal oxygen diffusion: high oxygen flow resolution: ± 1 pmol O₂ / s/mL from normoxia to anoxia; high oxygen concentration resolution: 5 nM. The glass chambers allow for working with 2 ml volumes, but 0.5 ml microchambers can be used to maximize sample concentrations and work with the smallest possible sample volume. The system maintains the temperature within a range of 4 to 47 °C with high stability and precision (Peltier thermostat). The equipment features PVDF stirrers with electromagnetic control to maximize the resolution of oxygen consumption measurements and includes software for data acquisition and analysis.
