The LSM900 inverted confocal optical microscope with Airyscan 2 offers excellent performance for acquiring high-quality, high-resolution images. It allows operation in confocal or super-resolution modes.

 

Some features of the equipment are detailed below:

  • Solid-state diode laser excitation lines: 405 nm, 488 nm, 561 nm and 640 nm

  • Colibri 5 LED fluorescent lighting system with RGB-UV modules

  • Objectives:

    90 HE LED including 425/30 (DAPI, Alexa 405, Hoechst and similar), 541/30 (GFP, Fluo 4, FITC and similar), 592/25 (Cy3, TRITC, DsRed and similar) and 709/100 (Cy5, Alexa 631, TOTO-3 and similar)

  • Detectors:
    It has two GaAsP detectors.
    The Airyscan 2 super-resolution detector has 32 concentrically grouped detection elements, increasing photon arrival and efficiency, increasing the signal-to-noise ratio between 4 and 8 times, and allowing a super-resolution of 120 nm.

  • High-resolution motorized XY stage and motorized Z-axis focusing with 10nm steps. Several universal mounting frames.

  • ZEN pro software with several associated measurement modules such as TimeLapse, Z-stack, Tiles, Shuttle&Find or Autofocus

  • The Airyscan 2’s Multiplex measurement mode allows you to obtain up to four super-resolution image pixels in a single sweep, thus significantly increasing acquisition speed.

Both this confocal microscope and the scanning electron microscope at IDiBE can share the same type of sample holders and use the same software, so correlative CLEM (Correlative Light-Electron Microscopy) can be easily carried out by superimposing and merging the information obtained by both techniques in the same area of ​​the sample
